Principle
Usesacombinationofenzymaticandgravimetricmethods.ThisprocedureisbasedonthemethodpublishedinOfficialMethodsofAnalysis,15thed.,AOAC,Arlington,VA,Vol.II,Sec.985.29,1105(1990).
Application
Thisassaydeterminesthetotaldietaryfibercontentoffoodusingacombinationofenzymaticandgravimetricmethods.Samplesofdried,fat-freefoodsaregelatinizedwithheatstableα-amylaseandthenenzymaticallydigestedwithproteaseandamyloglucosidasetoremovetheproteinandstarchpresentinthesample.Ethanolisaddedtoprecipitatethesolubledietaryfiber.Theresidueisthenfilteredandwashedwithethanolandacetone.Afterdrying,theresidueisweighed.Halfofthesamplesareanalyzedforproteinandtheothersareashed.Totaldietaryfiberistheweightoftheresiduelesstheweightoftheproteinandash.
